I know that you can go to http://www.flashvideodemos.com/ and get a flash video of an actor introducing your site.

My question is if I have access to chroma key and understand how to film the video, how hard is it to get my own custom made introduction onto my site?

The real secret in doing this correctly is to shoot in HD...even though full HD doesn't translate to web the quality is noticeably higher. Check out http://www.liveactor.com
I know they shoot with HD Cameras.

I'm not so sure about the HD part.... sounds more like an advertisement then technical advice.
The "real", real secret is to use a video editing program (like After Effects) that allows you to export the alpha channel (the transparent part) along with the regular RGB channels. So do your green screen as usual but don't put the weather map up behind the guy, just export the video without anything behind the speaker. Just be sure to export with alpha. With newer After Effects you can export directly to .flv, just be sure to encode the alpha channel also. Or, transcode the video with Flash 8 Video Encoder and be sure to open the "advanced" setting and click "encode alpha"
Then play the video in a .swf with wmode set to transparent.
